ARIPIPRAZOLE

 ARIPIPRAZOLE

BRAND NAME:- ABILIFY ,ARIPRA,ARILAN,BILIEF 10,15mg tabs, ARIVE 10,15,20,30mg tabs.

ARIPIPRAZOLE is an antipsychotics is unique in being a partial agonist at D2 and 5-HT 1À receptor, but antagonist at 5- HT2 receptor. The high affinity but low intrinsic activity of aripiprazole for D2 receptors impedes dopaminergic transmission by occupying a large fraction of D2 receptors but activating them minimally . It is not sedation, may even cause insomnia.

PHARMACODYNAMIC:-

Aripiprazole has high affinity for serotonin type 2 (5HT2), dopamine type 2 (D2), alpha1 and 2 adrenergic, and H1 histaminergic receptors. It also acts on a number of other receptors with lower affinity. The exact method by which aripiprazole's action on these receptors translates to a clinically relevant effect is not yet known.

MECHANISM OF ACTION:-

The antipsychotic action of aripipra byzole is likely due to the agonism of D2 and 5-HT1A receptors though the exact mechanism has not been defined. Some adverse effects may be due to action on other receptors. For example, orthostatic hypotension may be explained by antagonism of the adrenergic alpha1 receptors .

Aripiprazole is quite long - acting (t½ ~ 3 days) ; dose adjustment should be done after 2 weeks treatment. It is metabolized by CYP3A4 as well as CYP2D6 ; dose needs to be halved in patients receiving ketaconazole or quinidine , and doubled in those taking carbamazepine.

USED :-

used to treat certain mental/mood disorders (such as bipolar disorder, schizophrenia, Tourette's syndrome, and irritability associated with autistic disorder). It may also be used in combination with other medication to treat depression. Aripiprazole is known as an antipsychotic drug (atypical type).

used to treat schizophrenia in adults and children at least 13 year old.

SIDE EFFECTS:-

Dizziness, lightheadedness, drowsiness, nausea, vomiting, tiredness, excess saliva/drooling, blurred vision, weight gain, constipation, headache, and trouble sleeping may occur. If any of these effects last or get worse, tell your doctor or pharmacist promptly.

DOSE :-  DIRECT BY PHYSICIAN.

Chlorthalidone Uses, Side Effects, and More....

Chlorthalidone  Brand name:- Hythalton 50,100mg , Hydrazide, Thalizide ( 12.5, 25) . Chlorthalidone is a diuretic which lowers blood pressure by removing extra water and certain electrolytes from the body. Over time it also relaxes blood vessels and improves blood flow. Pharmacodynamic:- Not 🚫 available.  Pharmacokinetics:- Chlortalidone is slowly absorbed from the gastrointestinal tract after oral ingestion. It has a long half-life and therefore a prolonged diuretic action, which results in continued diuretic effects despite a skipped dose. This prolonged action of chlortalidone despite missing doses may account for the higher efficacy of chlortalidone compared to the shorter half-life medication, hydrochlorothiazide. Chlortalidone is eliminated from the body mostly by the kidney, as unchanged drug. Thus, in persons with diminished kidney function, the clearance of chlortalidone is reduced and the elimination half-life is increased. As with other thiazide diuretics, ch...

Chlordiazepoxide Uses, Side Effects, and More

Chlordiazepoxide Brand name:- Librium 10, 25 mg tabs; Equilibrium 10mg tab. Chlordiazepoxide is a benzodiazepine. It works by increasing the action of a chemical messenger (GABA) which suppresses the abnormal and excessive activity of the nerve cells in the brain. Pharmacodynamic:-  Chlordiazepoxide has antianxiety, sedative, appetite-stimulating and weak analgesic actions. The drug seems to block EEG arousal from stimulation in the brain stem reticular formation. The drug has been studied extensively in many species of animals and these studies are suggestive of action on the limbic system of the brain, which recent evidence indicates is involved in emotional responses. Hostile monkeys were made tame by oral drug doses which did not cause sedation. Chlordiazepoxide revealed a "taming" action with the elimination of fear and aggression. The taming effect of chlordiazepoxide was further demonstrated in rats made vicious by lesions in the septal area of the brain. The drug dos...
