Chlordiazepoxide Uses, Side Effects, and More

 Chlordiazepoxide

Brand name:- Librium 10, 25 mg tabs; Equilibrium 10mg tab.

Chlordiazepoxide is a benzodiazepine. It works by increasing the action of a chemical messenger (GABA) which suppresses the abnormal and excessive activity of the nerve cells in the brain.

Pharmacodynamic:-

Chlordiazepoxide has antianxiety, sedative, appetite-stimulating and weak analgesic actions. The drug seems to block EEG arousal from stimulation in the brain stem reticular formation. The drug has been studied extensively in many species of animals and these studies are suggestive of action on the limbic system of the brain, which recent evidence indicates is involved in emotional responses. Hostile monkeys were made tame by oral drug doses which did not cause sedation. Chlordiazepoxide revealed a "taming" action with the elimination of fear and aggression. The taming effect of chlordiazepoxide was further demonstrated in rats made vicious by lesions in the septal area of the brain. The drug dosage which effectively blocked the vicious reaction was well below the dose which caused sedation in these animals.

Pharmacology:-

Chlordiazepoxide acts on benzodiazepine allosteric sites that are part of the GABAA receptor/ion-channel complex and this results in an increased binding of the inhibitory neurotransmitter GABA to the GABAA receptor thereby producing inhibitory effects on the central nervous system and body similar to the effects of other benzodiazepines.Chlordiazepoxide is an anticonvulsant.

There is preferential storage of chlordiazepoxide in some organs including the heart of the neonate. Absorption by any administered route and the risk of accumulation is significantly increased in the neonate. The withdrawal of chlordiazepoxide during pregnancy and breast feeding is recommended, as chlordiazepoxide rapidly crosses the placenta and also is excreted in breast milk.Chlordiazepoxide also decreases prolactin release in rats. Benzodiazepines act via micromolar benzodiazepine binding sites as Ca2+ channel blockers and significantly inhibit depolarization-sensitive Calcium uptake in animal nerve terminal preparations.Chlordiazepoxide inhibits acetylcholine release in mouse hippocampal synaptosomes in vivo. This has been found by measuring sodium-dependent high affinity choline uptake in vitro after pretreatment of the mice in vivo with chlordiazepoxide. This may play a role in chlordiazepoxide's anticonvulsant properties.

Pharmacokinetics:-

Chlordiazepoxide is a long-acting benzodiazepine drug. The half-life of chlordiazepoxide is from 5 to 30 hours but has an active benzodiazepine metabolite, nordiazepam, which has a half-life of 36 to 200 hours. The half-life of chlordiazepoxide increases significantly in the elderly, which may result in prolonged action as well as accumulation of the drug during repeated administration. Delayed body clearance of the long half-life active metabolite also occurs in those over 60 years of age, which further prolongs the effects of the drugs with additional accumulation after repeated dosing.

Despite its name, chlordiazepoxide is not an epoxide.

Mechanism of action:-

Chlordiazepoxide binds to stereospecific benzodiazepine (BZD) binding sites on GABA (A) receptor complexes at several sites within the central nervous system, including the limbic system and reticular formation. This results in an increased binding of the inhibitory neurotransmitter GABA to the GABA(A) receptor. BZDs, therefore, enhance GABA-mediated chloride influx through GABA receptor channels, causing membrane hyperpolarization. The net neuro-inhibitory effects result in the observed sedative, hypnotic, anxiolytic, and muscle relaxant properties.

Used:-

Used to relieve symptoms of anxiety, including nervousness or anxiety that happens before a surgery. It may also be used to treat symptoms of alcohol withdrawal. Chlordiazepoxide is a benzodiazepine.

Side effects:-

Confusion, Dizziness, Fatigue, Sedation, Sleepiness, Unsteadiness, Abnormality of voluntary movements, Balance disorder (loss of balance).

Dose:-  25-100mg , as directed by your doctor.

BOTULINUM TOXIN Uses, Side Effects, and More

BOTULINUM TOXIN ( A or B ) BRAND NAME:- BOTOX ,DYSPORT  BOTULINUM TOXIN  A and B are highly potent exotoxins produced by Clostridium botulinum that are responsible for ' botulism' ( a type of food poisoning). These neurotoxic proteins cause long- lasting loss of cholinergic transmission by interacting with axonal proteins involved in exocytotic release of Ach. Localized injection of minute quantity of botulinum toxin.  PHARMACODYNAMIC :- Botulinum toxin A inhibits the release of acetylcholine, relieving muscle contraction and spasm associated with many conditions, such as incontinence and dystonia. Cosmetically, botulinum toxin A paralyses muscles in the face to temporarily treat wrinkles. The maximum effects of muscle paralysis occur four to seven days after a dose. When injected at therapeutic doses, botulinum toxin A causes partial chemical denervation of muscle tissue, causing local reduction of muscle activity. Muscle atrophy may result, axonal sprouting may beg...

Clonazepam used, Side effects, Dose, & more

Clonazepam  Brand name:- Lonazepam , Clonapax , Rivotril 0.5, 1.0, 2.0 mg tab. Clonazepam is a benzodiazepine. It works by increasing the action of a chemical messenger (GABA) which suppresses the abnormal and excessive activity of the nerve cells in the brain. Pharmacodynamic:- The pharmacodynamic properties of clonazepam are common among benzodiazepines and include anticonvulsive, sedative, muscle relaxing and anxiolytic effects . Animal data and electroencephalographic investigations in man have shown that clonazepam rapidly suppresses many types of paroxysmal activity including the spike and wave discharge in absence seizures (petit mal), slow spike wave, generalized spike wave, spikes with temporal or other locations, as well as irregular spikes and waves Label . Moreover, the agent can also decrease the frequency, amplitude, duration, and spread of discharge in minor motor seizures Label . Generalized EEG abnormalities are more readily suppressed by clonazepam than are focal ...

CLOMIPHENE CITRATE Uses, Side Effects, and More

CLOMIPHENE CITRATE BRAND NAME:- CLOMIPHENE, FERTOMED, CLOFERT, CLOME 25,50,100mg tab.  Clomiphene is a selective estrogen receptor modulator (SERM) which treats female infertility. It works by promoting the release of reproductive hormones. This helps stimulate the release of eggs from the ovary (ovulation). PHARMACODYANIMCS:- Clomifene (previously clomiphene) is an orally administered, non steroidal, ovulatory stimulant that acts as a selective estrogen receptor modulator (SERM). Clomifene can lead to multiple ovulation, and hence increase the risk of conceiving twins. In comparison to purified FSH, the rate of ovarian hyperstimulation syndrome is low. There may be an increased risk of ovarian cancer and weight gain. Clomifene is capable of interacting with estrogen-receptor-containing tissues, including the hypothalamus, pituitary, ovary, endometrium, vagina, and cervix. It may compete with estrogen for estrogen-receptor-binding sites and may delay replenishment of intracel...
