Centchroman is a birth control pill. It acts by preventing the implantation of the fertilized ovum to the womb (uterus).

Pharmacodynamic:-


Centchroman (Ormeloxifene) is a selective estrogen receptor modulator (SERMs) with contraceptive activity. Also, it has been suggested that it may be beneficial in the treatment of breast cancer, osteoporosis, dermatitis, restenosis, endometriosis and uterine fibroids.

 Pharmacology:-

Centchroman ( ormeloxifene) is a selective estrogen receptor modulator (SERM). In some parts of the body, its action is estrogenic (e.g., bones), in other parts of the body, its action is antiestrogenic (e.g., uterus, breasts). It causes an asynchrony in the menstrual cycle between ovulation and the development of the uterine lining, although its exact mode of action is not well defined. In clinical trials, it caused ovulation to occur later than it normally would in some women, but did not affect ovulation in the majority of women, while causing the lining of the uterus to build more slowly. It speeds the transport of any fertilized egg through the fallopian tubes more quickly than is normal. Presumably, this combination of effects creates an environment such that if fertilization occurs, implantation will not be possible.

 Mechanism of action:- 

Ormeloxifene has both estrogenic and anti-estrogenic activity. As a contraceptive, ormeloxifene inhibits endometrial receptivity to blastocyst signals. This mechanism inhibits implantation without affecting nidatory estrogen and progesterone, the hypothalamo-pituitary-ovarian axis, follicle maturation, ovulation, mating behavior, gamete transport or fertilization, and the preimplantation development of embryos.

Used:- 

Used for contraception. It is also used in the treatment of dysfunctional uterine bleeding.

Side effects:-

Delay in menstrual cycle in some women,15 but this occurs in around 8% of users and usually in the first three months. The periods tend to settle down to a rhythm once the body gets used to the drug. Periods can get scanty over time in some women.

 Dose:- 

For contraceptive - 30 mg twice a week for 12 weeks followed by once a week. 

For dysfunctional uterine bleeding - 60 mg twice a week for 12 weeks, then once a week for 12 weeks.
